Single-cell RNA-seq reveal lineage formation in human preimplantation embryos

Mouse studies have been instrumental in forming our current understanding of early cell-lineage decisions in mammals, however similar insights into early human development are severely limited. To generate a comprehensive transcriptional map of human embryo formation we sequenced the transcriptomes of 1,469 individual cells from 79 human embryos during preimplantation development. Analyses of these data revealed when and how the first cell-lineages differentiate, and that the segregation of the trophectoderm, epiblast and primitive endoderm lineages are established in concert and coincide with blastocyst formation.
